Summary
Abstract: This data set describes the research carried out from 1988 to 2003 on samples
collected in the sea (Terra Nova Bay and Wood Bay), in several lakes (Edmonson
Point, Markham Island, Inexpressible Island, Teall Nunatak, Tarn Flat, Skua
Lake, Carezza Lake and Gondwana Lake) and in some terrestrial areas, during
four Italian Antarctic Expeditions (1988/89, 1992/93, 1994/95, 1998/99). The
preserved freshwater and sea-water samples were investigated at LM, SEM and
flow citometry and as regards the sea environment, the same surveys were
carried out also on sea-ice, on the underlying water column and on the melt
water samples. Moreover, from in vivo samples, collected in these same
environments, monospecific microalgal cultures were set up to ultrastructural,
biochemical, physiological and molecular surveys. The results obtained till now
are interesting as evidenced from the scientific papers.

Quality
A collection of marine (sea water and ice-sea), freshwater and terrestrial
microalgae from Ross Sea and several Victoria Land localities. Observation at
LM, SEM and flow cytometer of the samples collected and appropriately
preserved. Isolation of microalgae from natural samples through the growing on
selected culture media (F2, BG11, BM, BBM, Bristol). Pigment analyses by
spectrophotometry and HPLC. Photosynthetic apparatus analyses using PAM. DNA
extraction and amplification of some nuclear (18S rDNA, ITS) and plastidial
(rbcL, psaA) genes.Study on the PSII will be carried out using as model
organism the cyanobacterium Synechocystis PCC 6803 wild type and some mutants
strains for the H proteins which have been constructed and characterized in our
laboratory. The photooxidative damages on the PSII proteins and the ability of
this organism to repair the damage will be estimated; the cab gene family,
codifying for the proteins of the PSII,of Koliella antarctica will be used to
verify the cold adaptation mechanisms. These genes will be compared to similar
genes of Clamydomonas reinhardtii to verify the structural changes of the PSII
antenna of the Antarctic organism that seems to have a dissipative function.
