Summary
Abstract: ENGLISH

Since the crucial role having by the microorganisms in the nutrient cycling of
the coastal marine ecosystem in Antarctica, the aim of this project is to
analyze the effect of the incident UV radiation (enhanced by the stratospheric
ozone layer) on the Antarctic marine bacterial flora. Response of isolated
strains and natural communities obtained from different water column levels
will be analyzed in Potter Cove, Antarctica. In this instance of the study we
will use interferential filters in order to fractionate the previously studied
UV-A and UV-B fractions in more narrows bands. In addition, the role of the
water column as a natural factor of solar radiation attenuation will be
analyzed using fixed and dynamics systems that permit us a better simulation of
the vertical mixing regime. Bacterial viability, DNA damage and induction of
oxidative stress-related enzymes will be evaluated. Field experiments will be
complemented and compared with laboratory irradiation assays under controlled
conditions. Due to that the standards methods of bacterial taxonomy are
inadequate for environmental strains, we will use 16S rDNA sequencing to
classify the isolates. Changes in the structure of bacterial communities will
be inferred using RFLP techniques and, if it is possible, by DGGE and SSCP

Dado el papel fundamental que los microorganismos cumplen en el ciclo de los
nutrientes del ecosistema marino costero antartico, este proyecto tiene por
objetivo analizar el efecto que la radiacion UV incidente en Antartida
(incrementada por el agujero de ozono estratosferico) tiene sobre la flora
bacteriana marina. Se analizara la respuesta de cepas aisladas y comunidades
naturales de distintas profundidades a diferentes regimenes de radiacion UV, en
Caleta Potter, Antartida. En esta etapa se utilizaran filtros interferenciales
que permitan desdoblar, las ya estudiadas fracciones UV-A y UV-B en bandas
menores. Adicionalmente se estudiara el efecto de la columna de agua como
filtro natural de la radiacion, utilizando sistemas estaticos y sistemas
dinamicos que reflejen mejor el regimen de mezcla vertical natural. Se
evaluara la viabilidad, el dano al ADN y la induccion de enzimas de estres
oxidativo. Los ensayos a campo se complementaran y compararan con ensayos de
irradiacion en laboratorio. Dado que los metodos convencionales de
clasificacion bacteriana son poco satisfactorios con cepas ambientales, se
utilizara la secuenciacion de ADNr 16S para tal fin. Ademas, los cambios en la
estructura de las comunidades bacterianas se inferiran mediante tecnicas de
RFLP y, en lo posible mediante DGGE o SSCP.
